Summary[edit] DescriptionAPI English: ER morphology in E. histolytica. Trophozoites expressing GFP-KDEL-FLAG were seeded on glass bottom culture dishes (MatTeck) embedded in a collagen matrix and incubated at 37°C. Immediately before imaging a coverslip was added directly to the top of the matrix. The amoebas were then imaged using the Perkin Elmer Spinning Disc Confocal Microscope. All images were acquired using the 40× objective and taking one stack every minute for 1 h using a 0.5 µm Z-step.
